Cryopreservation of lines is accomplished by freezing sperm or embryos.
For optimal sperm cryopreservation, males mice aged between 6 weeks and 6 months, and having produced at least one litter are recommended. Two males of the same line usually give 10 frozen sperm straws.

For your research projects or to validate the quality of frozen sperm with a view to re-establishing a lineage, the platform offers you a service for the validation of the sperm fertilization capacity.
The validation is carried out on one hand with the analysis of the motility of the sperm after thawing, and on the other hand by an in vitro fertilization test (IVF).
Depending on your needs, it is possible to analyze the quality of embryos obtained by IVF using two methods:
- 2-cell embryo formation or,
- Birth of viable individuals (reimplantation in pseudogestant females).

General questions

Yes, you must have a valid ethics certificate number or in the case of an active protocol, have made the necessary changes with the institution’s ethics committee.

Transgenic animals or cells production
Yes, however for random injection, a construct/tyrosinase co-injection is strongly recommended to facilitate early identification of genetically modified animals.
For CRISPR projects, we recommend the use of the Easi-CRISPR system (Miura et al. Nature protocols. 2017) since its effectiveness has already been demonstrated. In addition, the Cas9 protein and TracrRNA are already in our possession.
If, however, you insist on using your equipment, you must provide a report demonstrating the purity of the material to be micro-injected (D.O. report 280/260, 260/230; picture of the gel). The quantity and concentration of the material to be micro-injected will be determined after discussion with a manager of the platform.
We routinely use the FVB mouse line. For all other lines, please contact us.
We micro-inject 100 to 150 embryos per session. Of these, 70 to 100 survive and are reimplanted into pseudogestant females.
Yes, it is possible to get an animal genotyping service generated through our genomics platform. Please see this platform’s page for details of this service.
If you would like to do the genotyping on your own, we will take the tissue samples. As the samples are sent at the customer’s expense, it will be our pleasure to send you the samples by the carrier of your choice.
As soon as the genotyping results are provided and the animals are in good state of health, they will be available.
For animals to be moved to another institution, please contact us. Note that the transport costs will be the responsibility of the customer.
The process of creating founder mice takes a minimum of 10 weeks when there is no waiting. The order of service requests is based on the construction arrival dates.
Cryopreservation of lines
For each line to be cryopreserved, it is necessary to provide the males. The number of animals to provide will be determined according to your project. For mice, males should be between 2 and 6 months old and have already produced at least one weaned litter each.
WARNING. Reception of animals from another institution is subject to the approval of the UQAM veterinarian.
